Vitamin D2 vs D3: Which One Does Your Lab Report Show?
Your lab report shows "25-hydroxy Vitamin D: 22 ng/mL." Or sometimes: "25(OH)D: 22 ng/mL." Some reports show one number; others show D2 and D3 separately. What are these two forms, and does the distinction matter when you are supplementing?
Two Forms of Vitamin D
There are two natural forms of Vitamin D:
Vitamin D3 (cholecalciferol)
- Produced in the skin when exposed to UVB sunlight
- Found in animal-based foods: fatty fish (salmon, sardines, mackerel), egg yolks, liver
- The form used in most oral supplements in India
Vitamin D2 (ergocalciferol)
- Found in plant-based sources: UV-irradiated mushrooms
- Used in some fortified foods
- Also used in some prescription Vitamin D preparations
Both D2 and D3 are converted by the liver into 25-hydroxy Vitamin D (25(OH)D) - the form measured in blood tests. This is why the blood test is called "25-hydroxy Vitamin D" or "25(OH)D," not simply "Vitamin D."
What Your Lab Report Shows
Most routine Vitamin D tests measure total 25(OH)D - the sum of D2 and D3 forms. This is what the number on your standard report represents.
Some labs break it down:
- 25(OH)D2 (from plant sources or D2 supplements)
- 25(OH)D3 (from sun exposure, animal food, or D3 supplements)
- Total 25(OH)D = D2 + D3
If you have been supplementing with a D2 preparation or recently eaten UV-treated mushrooms, your D2 component may be higher. Most people without specific plant-based supplementation have predominantly D3 in their blood.
Is D3 Better Than D2?
This has been a debated question. Current evidence suggests:
- D3 is more potent at raising serum 25(OH)D levels - it produces a larger increase per unit of supplementation compared to D2
- D3 has a longer half-life in the body - it is stored more effectively
- D2 and D3 are both effective at correcting deficiency when given in adequate doses; the difference becomes more meaningful with lower doses or intermittent dosing
Most Indian commercial Vitamin D supplements use D3. The common 60,000 IU cholecalciferol sachets widely prescribed in India use D3. If you are buying over-the-counter Vitamin D, check the label for "cholecalciferol" (D3) vs "ergocalciferol" (D2).
Vegetarian and Vegan Consideration
D3 is traditionally derived from lanolin (sheep wool fat) - making it not vegan and, in some interpretations, not vegetarian. D2 is plant-derived and vegan-friendly. Some brands now offer vegan D3 sourced from lichen - this is both vegan and retains D3's potency advantage.
If you are strictly vegetarian or vegan and supplementing, verify whether your supplement is D2 or lichen-derived D3.
